Abstract

In this study, we extracted polysaccharides from Ascidian tunic, and investigated to determine its physicochemical properties and antioxidant activities. The yields of hot water extract (HE), autoclave extract (AE), and enzyme extract (EE) were 7.30, 9.60, and 10.20%, respectively. The uronic acid and hexosamine contents of EE was highest of 7.25 g/100 g, 33.01 g/100 g respectively. The free amino acid contents of EE were higher than those of AE and HE. The oxygen radical absorbance capacity (ORAC), FRAP activity and ferrous ion chelating effect of EE were 364.02 ¥ìM/g FW, 506.09 ¥ìM, and 32.33%, respectively. DPPH radical scavenging activities, reducing power and tyrosinase inhibitory activity increased with an increase in extract concentration. Tyrosinase activity of EE improved with higher treatment concentration. The rate of inhibition of MMP-1 production in HS68 cells exposed to UVB was suppressed by treatment with EE were 75.11% at 100 ¥ìg/mL. In conclusion, we provided experimental evidence that extracted polysaccharides from Ascidian tunic have potential as functional materials.
